چکیده

When pigs are reared from birth on a milk diet deficient in copper they develop characteristic cardiovascular defects that usually terminate in rupture of the heart or major vessels at 3 to 4 months of age (1). These events are preceded by demonstrable reduction of tensile strength of the aortas (2). The aortic elastic tissue shows histological defects (l), and alterations of its mechanical properties have been demonstrated.1 A reduction in insoluble elastin residue and an increase in a sodium chloride-extractable macromolecular component of these aortas has been reported (4). This report concerns the relation of the extractable material to the major intercellular components of 6he aorta, viz. collagen, elastin, and ground substance.

Induction of increased collagen and elastin biosynthesis in copper-deficient pig aorta.

Young pigs raised on a copper-deficient diet develop severe abnormalities of connective tissue due to defective cross-linking of collagen and elastin. They eventually succumb to anemia and cardiovascular damage, the latter apparently due to the defective connective tissue metabolism. We evaluated the effects of nutritional copper deficiency upon collagen and elastin synthesis using short-term e...

Membrane and contractile properties of rat vascular tissue in copper-deficient conditions.

To determine the effects of copper deficiency on the vascular wall, female rats of the Wistar King A strain were fed a copper-deficient diet (0.58 ppm), and all delivered pups were kept on the same diet. The copper level of sera of all copper-deficient animals was significantly lower (10.6 fig/ dl) than that of the controls (141.4 jug/dl). The body weight of copper-deficient rats was about 40% ...

Tropoelastin production and tropoelastin messenger RNA activity. Relationship to copper and elastin cross-linking in chick aorta.

The elastin content of the chick thoracic aorta increases 2--3-fold during the first 3 weeks post-hatching. The deposition of elastin requires the covalent cross-linking of tropoelastin by means of lysine-derived cross-links. This process is sensitive to dietary copper intake, since copper serves as cofactor for lysyl oxidase, the enzyme that catalyses the oxidative deamination of the lysine re...

Carbohydrate content of insoluble elastins prepared from adult bovine and calf ligamentum nuchae and tropoelastin isolated from copper-deficient porcine aorta.

1. Insoluble elastin has been prepared by several different methods from adult bovine and calf ligamentum nuchae. Highly purified tropoelastin has been prepared from copper-deficient porcine aorta. 2. Amino acid analyses indicated that all preparations, except that obtained from calf ligamentum nuchae by using an EDTA extraction followed by collagenase digestion (preparation E6), were typical o...

Dichotomous mechanisms of aortic stiffening in high‐fat diet fed young and old B6D2F1 mice

Abstract Advancing age is associated with increased stiffness of large elastic arteries as assessed by aortic pulse wave velocity (PWV). Greater PWV, associated with increased risk of cardiovascular diseases, may result from altered expression of the extracellular matrix proteins, collagen and elastin, as well as cross-linking of proteins by advanced glycation end products (AGEs). Indeed, aorti...
